In some instances, grandparents may experience concerns regarding their biological relationship with a grandchild. To ascertain this relationship, they may choose to undergo a grandparent DNA test. This genetic analysis is designed to determine the probability of a biological connection between a grandchild and their grandparents, particularly in cases where the alleged father is unavailable for testing.

The process of grandparent DNA testing involves the collection of biological samples through buccal (cheek) swabs from the participating grandparent(s) and the grandchild. Within a laboratory setting, DNA is extracted from the collected cells, and specific genetic regions, known as short tandem repeats (STRs), are amplified using polymerase chain reaction (PCR) technology. The resultant DNA profiles of the grandparent and grandchild are subsequently compared, and a statistical analysis is conducted to generate a “grandparentage index,” which quantifies the likelihood of a biological relationship.
